Enhance enzyme production and prevent sluggish fermentation with X-SEED® Carbo Peptone

Nitrogen sources, such as yeast peptones, typically provide peptides that are metabolized not only to increase biomass and enzyme production but also to maintain homeostasis. 

In this study we explored the effects of a new type of peptone (X-SEED® Carbo Peptone), containing carbohydrates and short-chain peptides, on enzyme synthesis. Investigations were conducted using GFP-expressing Escherichia coli and Bacillus subtilis, two popular cell factories for industrial-scale enzyme manufacturing. 

Key observations

  • When replacing standard yeast peptone in rich complex media, X-SEED® Carbo Peptone enhanced GFP production in both E. coli and B. subtilis by 52% and 15% respectively. 
  • When used as a supplement in semi-defined mineral media, X-SEED® Carbo Peptone improved GFP production by 16% in E. coli cultures.
  • Following the same supplementation strategy, sluggish B. subtilis cultures were better able to kickstart cell growth, eventually translating into a 33% increase in enzyme production.
  • The data suggests that supplying a balanced combination of yeast-derived carbohydrates and peptides can significantly improve enzyme production.

*Biomass and fluorescence signals were obtained via online measurements in Biolector Pro. Experiments were performed in biological triplicates. Average values are shown, standard deviation of less than 5% was observed.
